  1. Can you tell us about your experience working in an animal research laboratory? Answer: In my previous role, I have gained extensive experience working in an animal research laboratory. I have been involved in various aspects of animal handling, including their care and welfare. I have performed procedures such as fear-conditioning, immunostaining, stereotaxic surgery, and sleep recording in rats and mice. I have a strong understanding of ethical considerations, standard operating procedures, and safety protocols related to animal research.
  1. Could you provide an overview of the DST-funded project, “Role of Conditioned Learning and Sleep in Adult Hippocampal Neurogenesis in the Rat”? Answer: The DST-funded project aims to investigate the role of conditioned learning and sleep in adult hippocampal neurogenesis in rats. The project seeks to understand how these factors influence the generation of new neurons in the hippocampus, a region of the brain associated with learning and memory. The project will involve conducting experiments, including fear-conditioning protocols, immunostaining techniques, and sleep recording. I would diligently follow the protocols and procedures outlined by DST, adhering to ethical considerations and legal requirements. Regular communication and collaboration with the project team and DST officials would also be crucial to stay updated on any changes or updates to the guidelines.
  1. Can you provide examples of your experience with fear-conditioning, immunostaining, stereotaxic surgery, and sleep recording in rats and mice? Answer: During my previous work, I have gained hands-on experience with fear-conditioning, immunostaining, stereotaxic surgery, and sleep recording in rats and mice. I have successfully conducted fear-conditioning experiments, which involve training animals to associate a specific stimulus with a fearful response. I am skilled in performing immunostaining techniques to visualize specific proteins or markers in brain tissue sections. I have also conducted stereotaxic surgeries, a precise technique for targeting specific brain regions. Additionally, I have experience in sleep recording using appropriate equipment and software to monitor and analyze sleep patterns in rodents.
  1. How do you ensure the welfare and ethical treatment of animals in an animal research laboratory? Answer: The welfare and ethical treatment of animals are paramount in an animal research laboratory.   1. How would you approach performing sleep recording in rats and mice? Answer: When performing sleep recording in rats and mice, I would first ensure a suitable experimental setup that provides a conducive environment for sleep monitoring. This includes appropriate bedding, temperature, lighting, and noise control. I would carefully place electrodes or sensors to capture sleep-related parameters, such as brain activity, electromyography (EMG), and electrooculography (EOG). I would monitor the animals during the recording period, ensuring their comfort and safety. Following the recording, I would analyze the sleep data using appropriate software and apply established scoring criteria to classify sleep stages accurately.
