Desirable: Experience of at least three years in mammalian cell cultures, biochemical techniques, mass spectrometry, and knowledge of glycobiology and host-pathogen interactions.
Job Description: To investigate glycosylation of CD antigens in mammalian cells, isolate, and characterize them using the glycoproteomic methodologies. Mammalian cell culture, western / lectin blotting, confocal microscopy, flow cytometry, peptide and glycopeptide synthesis, and bio-analytical techniques.
Emoluments – Rs. 35,000/- per month + 24% HRA (As per DST OM dated 30.01.2019)

Answer: I am well-versed with mass spectrometry techniques, particularly in the context of glycoprotein analysis. In one of my previous projects, we employed mass spectrometry to study O-glycosylation patterns of a specific protein, similar to the objectives of this project. We successfully identified glycopeptides and determined their glycosylation sites, which greatly contributed to our understanding of immunological processes.
- Question: Explain the importance of glycosylation in CD antigens and its relevance to immunological processes. Answer: Glycosylation plays a critical role in modulating the functions of CD antigens, influencing various aspects of immunological processes. It affects the stability, trafficking, and signaling properties of these cell surface proteins. By studying the glycosylation patterns of CD antigens, we can gain insights into their functional significance and their involvement in immune responses.
- Question: Have you worked with biochemical techniques like western blotting and lectin blotting? How have these methods contributed to your previous research? Answer: Yes, I have utilized biochemical techniques such as western blotting and lectin blotting in my previous research. These methods have been instrumental in characterizing glycoproteins, including CD antigens, by detecting glycan structures. By analyzing lectin binding patterns and western blot profiles, we could identify specific glycosylation modifications and understand their potential roles in cellular processes.
- Question: Can you explain the significance of peptide and glycopeptide synthesis in the context of this project? How can these techniques aid in glycosylation studies? Answer: Peptide and glycopeptide synthesis are essential for studying glycosylation events and mapping glycosylation sites. By chemically synthesizing specific peptides and glycopeptides, we can create reference standards for mass spectrometry analysis, facilitating the identification and quantification of glycosylation modifications. These synthesized molecules also enable us to investigate the impact of specific glycan structures on protein functions, providing valuable insights into the glycoproteomic profiling of CD antigens.
